Association of IFN (+874 T/A), IL-17 (-152 A/G) AND IL-4 (-590C/T) Polymorphism with Pulmonary Tuberculosis

Abstract
Introduction: Cytokines play an important role in determining the susceptibility or resistance to
intracellular pathogen Mycobacterium tuberculosis. The important cytokine IFN play a central
role in initiating a programmed mechanism to act against the tubercle bacilli. Other cytokines
such as IL-17, acts a synergy with IFN to acts against pathogen. IL-4 cytokines favor
intracellular survival of the bacilli. These cytokines production and action on bacilli in turn
depends upon the presence of single nucleotide polymorphism (SNP) of these individual genes.
In the present study we aim to determine the SNP of IFN (+874 T/A), IL-17 (-152 G/A), IL-4
(-590 C/T) in pulmonary tuberculosis patients compared with healthy controls.
Materials and Methods: A case-control study, genomic DNA was extracted from the peripheral
blood from the tuberculosis cases as well as from healthy controls. SNP of IFN (+874 T/A),
IL-17 (-152 G/A), IL-4 (-590 C/T) were investigated by PCR and RFLP. A total of 155 healthy
controls and 150 tuberculosis cases were included.
Results: The IFN frequent genotype found in cases and controls are TA and followed by AA
and TT, there is no significant between cases and controls. TA associated with TB cases
(OR-0.638,95%CI-0.376-1.081, P=0.108), The IL-17 (-152 G/A) polymorphism, AG and AA
genotype were more likely associated with tuberculosis, when compared to the GG genotype
(OR-1.968, 95% CI- 1.195-3,241, p=0.007). IL-4 (-590 C/T) genotypes, the heterozygote
genotype CT was statistical significantly higher in tuberculosis cases than controls (OR-4.475,
95%CI-2.626-7.626, p=0.00001). The gene combination study IFN TT hi IL-17 AG low
(OR-0.32, 95% CI-0.13-0.74, p=0.006) found more in healthy controls, The gene combination of
statistically significant combinations, IFN TA Int IL-4 CT hi (OR-7.11, 95%CI-4.03-12.56,
p=0.00) has been found to be associated with tuberculosis.
Conclusion: In conclusion, single nucleotide polymorphisms studied in different cytokines
IL-17, IL-4 has been found to be associated with the tuberculosis. Multi gene combination
studies helped us to find the person risk of developing tuberculosis.
